Lors du traitement de première ligne du carcinome mammaire métastatique, on a comparé l’administration de paclitaxel (220 mg/m²), effectuée 24 heures après l'administration de doxorubicine (50 mg/m²), au moyen d'une perfusion durant trois heures, à la thérapie standard FAC (5-FU : 500 mg/m², doxorubicine : 50 mg/m², cyclophosphamide 500 mg/m²). Par rapport à la thérapie standard, l’association paclitaxel/doxorubicine augmentait la fréquence et la sévérité des effets indésirables suivants : neutropénie, anémie, neuropathie périphérique, arthralgies/myalgies, asthénie, fièvre et diarrhée.
Bij de eerstelijns behandeling van gemetastaseerd mammacarcinoom werden neutropenie, anemie, perifere neuropathie, arthralgie/myalgie, asthenie, koorts en diarree vaker, en als zijnde ernstiger, gemeld wanneer paclitaxel (220 mg/m 2 ) 24 uur na doxorubicine (50 mg/m 2 ) werd toegediend als een 3- uurs infuus in vergelijking met standaard FAC therapie (5-FU 500 mg/m 2 , doxorubicine 50 mg/ m 2 , cyclofosfamide 500 mg/m 2 ).
